IndicationVelakast is a combination therapy and it contains two active ingredients: Sofosbuvir and Velpatasvir. Velakast is available in the form of tablets and strength of 400mg/100mg (Sofosbuvir 400mg + Velpatasvir 100mg). This antiviral medication is highly effective in the treatment of chronic HCV (hepatitis C virus) infection and it works by preventing the spread of the virus, which helps in decreasing the amount of infection from the body. Do not perform any activity that requires concentration and strength because the use of Velakast tablets may increase the risk of dizziness and loss of strength in some patients.
PharmacologySofosbuvir is the first active ingredient present in the Velakast and it is a nucleotide analog inhibitor that inhibits the RNA-dependent RNA polymerase of the HCV NS5B (non-structural protein 5B). Sofosbuvir is incorporated into HCV RNA by the NS5B polymerase and functions as a chain terminator Synthesis after intracellular metabolism to generate the pharmacologically active uridine analog triphosphate (GS-461203). Sofosbuvir inhibits HCV viral replication by binding to the two Mg2+ ions found in the GDD active site motif of the HCV NS5B polymerase and blocking the replication of HCV genetic material. Velpatasvir is the second active ingredient present in the Velakast and it is a small molecule direct-acting antiviral used in the treatment of hepatitis C in combination with sofosbuvir. Velpatasvir prevents viral replication by inhibiting NS5A (non-structural protein 5A).NS5A inhibitors compete with RNA for binding. NS5A inhibitors are also thought to bind the target during replication when the binding site is revealed. Inhibition of NS5A is also known to cause the protein to redistribute to lipid droplets.
